In August of 1814, the village of Loudonville was laid out by Stephen Butler and James Loudon Priest. Stephen Butler had lived in the village before it was laid out, and his home became a tavern and hotel. The rest of the village slowly built up around it. Loudonville's post office was first erected in 1820. The first bank in the area was built in 1868. A large fire broke out in 1901, causing massive damage and the destruction of several buildings. Loudonville residents took this opportunity to improve the town.

Starting in 1913, the Flxible Company called Loudonville home until it closed down in 1996. This American manufacturer produced transit buses, funeral cars, motorcycle sidecars, and other commercial vehicles for decades. It even produced various war goods during World War II. Sections of the factory were still in use by Motor Coach Industries until the mid-2010s.

Loudonville is home to the 4,525-acre Mohican-Memorial State Forest and the 1,110-acre Mohican State Park. There are also three nationally recognized historic sites in the city. One of the most distinguished local landmarks is the Ohio Theatre.

The town's first theatrical building was built in 1874 but was eventually demolished, allowing the current Ohio Theatre to be constructed in its place in 1910. This new building housed offices, council chambers, and even a library. In the 1990s, volunteers with the Mohican Area League of Arts helped to restore the establishment. It was placed on the National Register of Historic Places in 1997 as the City Hall and Opera House. The theater is still in operation today and often partners with local schools for student performances.
